Ipamorelin (NNC 26-0161) is a selective growth hormone secretagogue — a pentapeptide agonist of the ghrelin / growth hormone secretagogue receptor (GHS-R1a). It is frequently described in the literature as the first GH secretagogue to release growth hormone with high selectivity, meaning that in the foundational research it stimulated GH without meaningfully raising ACTH, cortisol, or prolactin — a profile that distinguishes it from earlier secretagogues such as GHRP-2 and GHRP-6.

Research Context & Mechanism+
Ipamorelin binds the growth hormone secretagogue receptor (GHS-R1a), the same receptor targeted by the endogenous hormone ghrelin. Activation at the pituitary drives release of stored growth hormone in a pulsatile fashion. Because it acts as a secretagogue — prompting release of the body's own GH rather than supplying GH directly — it is studied as a tool compound for probing the GH axis.
Its defining characteristic in the literature is selectivity. In the foundational characterization by Raun and colleagues (1998), Ipamorelin released growth hormone with a potency comparable to GHRP-6, but — unlike GHRP-6 and GHRP-2 — did so without significant elevation of ACTH or cortisol, and without meaningfully affecting prolactin, FSH, LH, or TSH. This "clean" GH-selective profile is the reason it is often referenced as the first selective GH secretagogue.
Beyond the GH axis, Ipamorelin has been examined in gastrointestinal-motility research as a ghrelin-mimetic, including rodent models of postoperative ileus (gastric dysmotility).
Selectivity vs Other Secretagogues+
A common reason Ipamorelin appears in comparative research is its narrow hormonal footprint relative to earlier growth hormone-releasing peptides:
- GHRP-6: reported in the source literature to release GH, with concurrent orexigenic activity and elevation of cortisol/ACTH.
- GHRP-2 (pralmorelin): potent GH release, but with more pronounced ACTH, cortisol, and prolactin effects.
- Ipamorelin: GH release comparable to GHRP-6 in the original work, with ACTH and cortisol reported at or near baseline.
It is frequently studied alongside the GHRH analog CJC-1295, because a GHRH analog plus a GHS-R agonist act on two complementary pathways of the GH axis.
Development Status & Honest Limits+
Ipamorelin was originally developed by Novo Nordisk (as NNC 26-0161) and later investigated by Helsinn Therapeutics in Phase II trials for postoperative ileus. That clinical program was discontinued for lack of efficacy on its primary endpoint, and Ipamorelin has never been approved as a therapeutic drug. Most of the human GH-release data comes from small early-phase pharmacology studies rather than large controlled trials.
We flag this deliberately: the selectivity findings are well characterized in preclinical and early clinical work, but Ipamorelin remains an investigational research compound, not an approved medicine.
